Amorolfine nail lacquer is a generic treatment used to medicate fungal nail infections. The branded version of the treatment is called Loceryl, and is marketed by Galderma. It is applied directly to the affected fingernails or toenails, normally on a once- or twice-weekly basis for a period of several months.

Fungal nail infections are caused by an imbalance of fungi on the fingernails or toenails. This in many cases is the result of a fungal infection called athlete’s foot, spreading to the nail from surrounding skin. Several different varieties of fungi live on the skin, usually harmlessly. Under certain conditions however, some types of fungi can become more prominent, with their cells multiplying and spreading. This can upset the balance of skin fungi and lead to an infection.

The main cause of nail infections are a type of fungi called dermatophytes, however on some occasions the fungi Candida can them too. These type of fungi tend to thrive in warm, damp, dark conditions; so people who wear tight shoes or spend a lot of time with wet or sweaty feet tend to be more susceptible to them. People who have certain medical conditions affecting their circulation or immune system, such as diabetes, are also more likely to get fungal nail infections.

There are measures that can be taken to reduce the likelihood of a fungal nail infection occurring, such as wearing footwear that fits properly, and keeps the feet dry. People undertaking sports or physical activity are also advised to use footwear that allows the feet to ‘breathe’ so their feet don’t become too damp with sweat.

It’s rare for complications to develop as a result of a fungal nail infection, but they can be unsightly in appearance and cause discomfort. They can also spread to other nails and to other people; so getting treatment for them is recommended.

The active ingredient in Amorolfine nail lacquer is an antifungal by the same name. To survive and spread, fungal cells need to produce a protein called ergosterol. This helps the cell to maintain its structure. Antifungals such as amorolfine work by preventing the fungus from producing this protein. This then causes holes to appear in the walls of the fungal cell, and the contents leak out. Following this, the fungal cells die, and this helps to neutralise the infection.

Directions

Follow the instructions given by your prescriber when using this medication, and make sure you read the patient information leaflet prior to use.

  • The typical dose is one application once or twice weekly to the affected finger or toe nail(s). Your doctor will provide instructions specific to your symptoms.
  • Using the nail file supplied, file down the infected parts of the nail and nail surface as much as you can. Do this for each affected nail.
  • Make sure you do not use this nail file on healthy nails, or you may spread the infection.
  • Wipe the infected nails with the swabs supplied so they are clean.
  • Dip the applicator into the lacquer solution, without wiping off any excess when pulling it out of the bottle.
  • Apply evenly over the whole of the affected nail. Repeat this for any other affected nails.
  • Give the lacquer around three minutes to dry.
  • Clean the applicator following use.
  • For subsequent applications, make sure you clean off any of the previous lacquer with one of the swabs provided before administering a new layer, and again file down the infected parts of the nail beforehand if necessary.
  • Treatment may need to continue for 6 months for fingernails, or for up to 9 or 12 months for toenails.
  • Do not use more than the recommended dose.
  • Do not swallow the lacquer. Seek immediate medical attention if you or anyone else accidentally does so.

Side effects

If you experience any signs of an allergic reaction when using this treatment, such as hives, difficulty breathing or swelling of the lips, face or throat, seek medical attention immediately.

Rare (1 in 1000 people or less):

Discoloration of the nail, loosening of the nail from the nail bed.

Very rare (1 in 10,000 people or less):

Burning sensation or allergic skin reaction.

This medication is not thought to interact with alcohol.

In a safe place, out of the reach of children, with the bottle sealed firmly tight and under 30°C.

Am I allergic to anything in the medicine?

This lacquer contains: amorolfine, Eudragit RL 100 (ammonio methacrylate copolymer A), triacetin, butyl acetate, ethyl acetate and ethanol (anhydrous).

Do not use it if you are allergic to any of the above.

Is it available over-the-counter?
